As John mentioned in an earlier post you may have what Keystone calls "racetrack ducting". In other words an endless loop so blocking the end outlet will defeat the design. Check the brochure for your year and model. YMMV

The vent is round and rotates??? If so, use your fingers and "snap" it off at the edge, it will fall off into your hand. To reinstall, just "snap it back in place".

I would not use spray foam. It expands and may, if you misjudge the amount or location, expand to split the duct or creep into an area you don't want foam to seal. I'd suggest either a small piece of closed cell foam, a $1 swim noodle or something similar. Cut it to shape and slightly larger than needed so it will "wedge into place" and then push it into the duct carefully. If you ever do need to remove it, it's much easier to get that kind of "block" out of the duct. You'll NEVER be able to cut away expanding foam once it sets up, even if it is in the wrong place and must be removed.....

Hi the a/c race track in our 2019 Cougar RLS ends in the bedroom sky lite, where you would install the 2nd a/c. I opened it up because there was more cold air coming out around the sky lite vent instead of the a/c vents. Found a very lousy tape job on the ends(imagine that) I used a cut pool noodle and some HVAC tape to seal it off.Now the whole system works much better.

You can also use a camera phone to take a picture down the duct

Can also put the top end of your cell phone up into the duct and take a picture using the flash.
