Welcome to the forum. Let's begin with a model number for that Premiere. Saying you have a 2020 Premiere is like saying you have a 2020 Chevy truck. If you go to the UserCP and add a signature with the year, make, and full model info of trailer and truck it will show with every post.
Now on to your issue. Was the tank gate valve working previously or is this a first trip with a new to you camper? "After 2 weeks of camping", most folks can't go 2 weeks without dumping their tanks. Not knowing what model you have I have no way of knowing the blank tank capacity. Was the valve left open? How did you determine the tank is full? Did it back up into the toilet or are you relying on the tank level indicator lights?
As for the valve being difficult to open, more questions, sorry but it requires more information to make a relevant suggestion. Does the handle NOT move to open the valve or does it open the valve but it's very stiff and difficult? Again, without knowing a model number I don't know if that valve is cable operated or directly connected with an extension rod. Cable operated valves can get kinked if they aren't secured. If the valve isn't opening then dropping the belly to get to the valve is inevitable.
If the valve isn't opening then adding an enzyme like RV Dissolve won't fix the mechanical issue.The enzymes take a while (days or weeks) to work so you won't see immediate or near term results..
