Wood rot under roof?

I recently discovered a tear on my EPDM roof. Seeing as how I just found it and its been sitting all winter, it has been there for at least 6 months.

I live where it rains all the time so I want to patch it with Eternabond but since it has been like this for a while, do I need to tear more of that rubber membrane up and see if that would is all water logged and rotting?

To give you an idea the size of the tear, a quarter could cover it all. Thanks

I might be inclined to cut a clean line down the middle of the "puddle" looking section. That way you can assess the amount of moisture, deal with it and have a clean line to seal it back up.

Its weird that there are other sections that have that same look "puddle section" but I don't see any holes in other places. I ordered eternabond so when I get that I'm gonna cut a section and see what the wood looks like.
